ABSTRACT

Objective@#To observe the association between puberty development and thyroid function among school-aged girls in Minhang District of Shanghai, in order to explore the effect of puberty development on thyroid function. @*Methods@#The study was based on a cohort of adolescent girls recruited in iodine-suitable areas of Minhang District, and the baseline and follow-up survey have been carried out from January to March 2019. The method of phased cluster sampling was used to select one junior high school in the east, south, north and middle of Minhang District, Shanghai, respectively. Finally, 464 new junior high school girls were included in the Cohort study for physical examination, and girls were followed up from January to March 2021. The Puberty Development Scale (PDS) was used to assess the stage of puberty. Serum thyroid stimulating hormone (TSH), free triiodothyronine (FT3), and free thyroxine (FT4) were determined for each participant. Thyroid homeostasis structure parameters (THSPs) was calculated. Changes of Thyroid hormones at baseline and follow-up were compared by Wilcoxon signed rank sum test. Multiple linear regression analyses were used to evaluate the associations of thyroid hormones (THs) and THSPs changes with pubertal develepment.@*Results@#Serum TSH levels of female studentds decreased significantly, while their FT3 and FT4 levels increased significantly during the study period ( Z=-10.53, -4.71, -12.46, P <0.01). In multiple linear regression analysis after adjustment for co-variables (including baseline age, change of BMI and waist circumference), FT4 and thyroid feedback quantile-based index (TFQI) in the higher puberty category scores changes (△PCS) group were further reduced compared with those in the low △PCS group ( β =-0.66, -0.55 ). Compared to the late puberty at baseline and follow-up (BLFL) group, FT4 and TFQI showed higher decline in the pre-puberty at baseline and late puberty at follow-up (BPFL) group with the pre-puberty at baseline and end of puberty at follow-up (BPFT) group ( β =-0.55, -0.44)( P <0.05). There were no association of △TSH, △FT3, △FT4/FT3 and TSH index changes (△TSHI) with △PCS or the puberty pattern.@*Conclusion@#Serum TSH decreases while serum FT3 and FT4 increase among girls during puberty. Both the initial stage and the velocity of pubertal development are related to thyroid hormone fluctuations.

ABSTRACT

OBJECTIVE: To explore the situation of knowledge-attitude-practice of medication (shorted for medication KAP) of elderly patients with chronic diseases and its relationship with depression mood. METHODS: Through using scale of medication KAP in elderly adults with chronic diseases and Self-rating Depression Scale (SDS), a face to face questionnaire survey was conduceed among elderly patients with chronic diseases examined in two community service centers of Lanzhou. The Pearson correlation analysis was used to verify the correlation between the score of medication KAP and the score of depression mood, and multivariate stepwise linear regression was used to analyze the influential factors (including the items in the general data table and depression mood with statistical significance after single factor analysis) of the score of medication KAP. RESULTS: Totally 300 questionnaires were sent out, and 289 valid questionnaires were returned with effective feedback rate of 96.3%. The results of questionnaire investigation showed the average score of the KAP was (31.12±6.20); the scores of medication knowledge, attitude and practice was (18.39±4.85), (3.18±1.24) and (9.55±2.08), respectively. The average score of depression mood was (39.99±6.06), compared with 48.4% (140 patients) with depressive symptoms. Pearson correlation analysis showed that the score of medication knowledge (r=-0.356), the score of medication compliance (r=-0.465) and the score of medication KAP (r=     -0.490) were negatively correlated with depression mood (all P<0.01). Multiple stepwise regression analysis indicated that the score of the KAP were affected by depression mood and demographic factors including family occupation, detailed monthly income in a family, the difference was statistically significant(P<0.05). CONCLUSIONS: In the community, the level of the medication KAP in elderly patients is mainly affected by depression mood and also by demographic factors. In the work of health care education for elderly patients with chronic diseases, medical staff should pay more attention to patients with depression mood and timely take targeted nursing measures to improve the level of the medication KAP.

ABSTRACT

A quantitative nuclear magnetic resonance spectroscopic ( qNMR) method was established for the simultaneous determination of resveratrol and polydatin in Polygonum Cuspidatum traditional Chinese herb cuts and granule. The 2_step ultrasonic extraction method using 80% alcohol and acetone was used for fully extracting these two components in samples before qNMR determination. The qNMR experimental conditions were investigated and deuterated dimethyl sulphoxide_deuterium oxide (10∶1, V/V) was selected as solvent, the pulse delay time was 5 s, the scan number was 32, 2,3,5_triiodobenzoate was used as internal standard which was calibrated with primary standard substance of potassium hydrogen phthalate. The 1 H_NMR peaks atδ6. 388-6. 391 ( d, 2H) of resveratrol and δ 6. 322-6. 330 ( t, 1H) of polydatin were chosen as the quantitative peaks. Method validation was performed in terms of precision ( RSD0. 999), limit of detection (0. 23 g/L for resveratrol and 0. 24 g/L for polydatin) and limit of quantitation ( resveratrol 0. 69 g/L, polydatin 1. 57 g/L), recovery ( resveratrol 97. 7% -103 . 5%, RSD=2 . 4%, polydatin 94 . 5%-99 . 2%, RSD=1 . 6%, including the sample extraction and preparaton process) . The results showed the reliability of qNMR for traditional Chinese medicine assay. The resveratrol and polydatin in Polygonum Cuspidatum real cuts and granule samples were experimental determined as 3. 57-5. 69 mg/g and 12. 73-24. 07 mg/g, respectively.

ABSTRACT

In order to enhance the alkaline polygalacturonate lyase (PGL) productivity by Pichia pastoris, we developed a constant cell concentration culture strategy by methanol feeding (called as CCCM culture) used in the continuous cultures. We controlled reasonable cell concentrations in the bioprocess by different strategies of methanol feeding. Using this CCCM culture with DCW 75 g/L, we significantly enhanced the PGL productivity (Qv) and the average specific enzyme production rate (Qx) of PGL to 6.11 U/(mL x h) and 81.5 U/(g x h), increased by 42.1% and 191.2% than the fed-batch culture with high cell density, respectively. The final PGL activity was 441.9 U/mL. Moreover, the extracellular protease concentration is 1.9 mg/L and the cell viability is more than 94% after 120 hour induction. The results show that this new strategy is advantageous in reducing proteolytic degradation and enhancing cell viability.

ABSTRACT

Lyophylized antler powder was hydrolyzed by pepsin and trypsin separately and also simultaneously to give hydrolysates with special physical activities. Complete hydrolysis peptides with MW lower than 1 x 10(3) were collected for assay of angiotensin I-converting enzyme (ACE) inhibitory activity, antioxidant activity and proliferative activity toward UMR-106 osteoblast cells. The results of the experiments revealed that all hydrolysates exhibited potent hydroxyl radical scavenging activity with an IC50 value less than 1 mg/ml which was much lower than the value of 5.5 g x L(-1) for vitamin C. The peptic and peptic tryptic hydrolysates demonstrated strong angiotensin I-converting enzyme (ACE) inhibitory activity. The tryptic hydrolysate increased the proliferation of the UMR-106 cells by 73.43%. The results verified the traditional use of antler in bone-strengthening, anti-aging. The exploratory studies on the ACE inhibitory activity of antler hydrolysates indicated that the hydrolysates might be potentially useful in prevention and treatment of hypertension. Further purification of peptides contributing to the antioxidant activity, angiotensin I-converting enzyme-inhibitory activity and proliferative activity toward osteoblasts from antler hydrolysates is warranted.

ABSTRACT

Objective To study the effect of intraoperative glucose and insulin infusion on the metabolism in patients undergoing esophageal cancer surgery under combined general anesthesia with epidural block. Methods Twenty ASA physical status Ⅰ-Ⅱ adult patients undergoing esophageal cancer surgery were studied. Ten patients received an iv glucose infusion at 0.5 g·kg~(-1)·h~(-1) and insulin infusion (1-1.5 IU ∶ 1 g glucose) throughout the anesthesia. Ten control subjects received isovolumic nutrient-free saline solution. Rectal temperature, plasma glucose concentrations, plasma free fatty acid (FFA) concentrations, plasma potassium concentrations and 24 hour urea nitrogen were measured perioperatively. Results No statistical difference was observed in rectal temperature and 24 hour urea nitrogen between the two groups. The plasma glucose concentrations continued to increase perioperatively in the control group. The glucose concentration increased during the operation in the glucose/insulin group, but the glucose concentrations at 1 and 2 hours after the operation were not statistically different from that before operation. Significant difference in plasma FFA concentration was found perioperatively (30, 90, 120, 150 min during the operation compared with 1 h after operation) between the two groups. In the glucose/insulin group, there was a significant decrease in the concentration of plasma potassium at 1 and 2 h during and after the operation. Conclusions Intraoperative glucose and insulin infusion cannot prevent hypothermia in patients undergoing esophageal cancer surgery under combined general anesthesia with epidural block, however, it may reduce lipoclasis.

ABSTRACT

<p><b>OBJECTIVE</b>To evaluate the feasibility of rapid prenatal diagnosis of chromosome aneuploidies by interphase fluorescence in situ hybridization (FISH) using uncultured amniotic fluid.</p><p><b>METHODS</b>Bacterial artificial chromosome (BAC) DNA probes were prepared and validated by using cultured peripheral blood. Interphase FISH for chromosomes 13, 18, 21, X and Y was performed in 60 amniotic fluid samples for the rapid prenatal diagnosis of chromosome aneuploidies, and the results were compared with the karyotypes from conventional cytogenetic analysis.</p><p><b>RESULTS</b>Of all 60 cases, 58 were concordant with their karyotypes, and 1 case of inv(9) and another case of t(2,12) were identified by karyotyping. Two cases of trisomy 21 and 1 case of trisomy 18 were detected by FISH and confirmed with conventional cytogenetics (sensitivity=100%). There were no false-positive or false-negative results.</p><p><b>CONCLUSION</b>This evaluation demonstrated that FISH employing BAC DNA probes could accurately and rapidly detect aneuploidies involving the above 5 chromosomes. However, as it does not identify structural chromosome aberrations and aneuploidies involving other chromosomes, it is not a substitute for conventional chromosome analysis, and the negative FISH result should be carefully interpreted.</p>
